Factors involved with cadmium absorption by a wild-type strain of Saccharomyces cerevisiae BJM

At the concentration used in this work (10 ppm), cadmium was efficiently removed from the environment by stationary yeast cells. While exponential phase cells showed low capacity of cadmium absorption, stationary cells removed 97% of the original metal in 24 hours. Total cadmium absorption shown by dry cells was lower than that of fresh ones, although both cells removed 50% of metal during the first hour of treatment. We also verified that only viable cells were capable of absorbing cadmium. Independently of the growth phase, cells showed high tolerance to 10 ppm CdSO4 and about 80% of cells remained viable after 24 hours exposure to cadmium. Abstract Polysaccharides from fungal wastes were partially characterized and evaluated for their protective effects against bacterial spot caused by Xanthomonas gardneri on four tomato cultivars: Santa Cruz Kada, Natália, BRS Sena and Forty. The polysaccharides were extracted from spent mushroom substrate of Pleurotus ostreatus, residual brewery yeast (Saccharomyces cerevisiae), and basidiocarps discarded from Lentinula edodes production. Fermentation capacity of Saccharomyces cerevisiae cultures BABT

This work aimed at the assessment of fermentative capacity of original diploid, monocellular haploid and recuperated diploid cultures of S. cerevisiae in sterilized sugar-cane wort. Twenty eight cultures were analyzed, four being original diploids (URM-4420, Itaiquara Ferment FIT, Lallemand Ferment FLA and Wild Ferment SEL); 12 monocellular haploids from original ones and 12 recuperated diploids from the monocells. The ethanol percentage ranged from 1.7 to 6.2% and the unfermentable reducing sugars from 0.45 to 0.50g/100mL. Double mutation of Saccharomyces cerevisiae for enhanced β-D-fructofuranosidase fructohydrolase productivity and application of growth kinetics for parametric significance analysis BJM

Abstract The kinetics of an extracellular β-D-fructofuranosidase fructohydrolase production by Saccharomyces cerevisiae in a chemically defined medium, i.e., sucrose peptone agar yeast extract at pH 6, was investigated. The wild-type was treated with a chemical mutagen, methyl methane sulfonate. Among the six mutants isolated, methyl methane sulfonate-V was found to be a better enzyme producing strain (52 ± 2.4a U/mL). The maximum production (74 ± 3.1a U/mL) was accomplished after at 48 h (68 ± 2.7a mg/mL protein). ABSTRACT The purpose of this study was to identify the yeasts involved in spontaneous fermentation of cocoa from the Brazilian Amazon region. The fermentation process was carried out experimentally with cocoa seeds from two sites (Medicilândia and Tucumã), State of Pará, northern Brazil, during a six-day period. Totals of 44 yeasts were isolated from Medicilândia and 29 from Tucumã. The Role of Tyrosine 207 in the Reaction Catalyzed by Saccharomyces cerevisiae phosphoenolpyruvate carboxykinase Biol. Res.

The functional signifcance of tyrosine 207 of Saccharomyces cerevisiae phosphoenolpyruvate carboxykinase was explored by examining the kinetic properties of the Tyr207Leu mutant. The variant enzyme retained the structural characteristics of the wild-type protein as indicated by circular dichroism, intrinsic fuorescence spectroscopy, and gel-exclusion chromatography. Kinetic analyses of the mutated variant showed a 15-fold increase in Km CO2, a 32fold decrease in Vmax, and a 6-fold decrease in Km for phosphoenolpyruvate. These results suggest that the hydroxyl group of Tyr 207 may polarize CO2 and oxaloacetate, thus facilitating the carboxylation/decarboxylation steps.

O objetivo deste trabalho foi avaliar o efeito de tratamentos pré-colheita sobre a ocorrência de podridões pós-colheita e sobre os atributos de qualidade de framboesas (Rubus idaeus L.) 'Heritage'. As frutas foram pulverizadas com um dos seguintes tratamentos: água destilada (controle), 6 g L-1 de quitosana, 100 mg L-1 de dióxido de cloro, Bacillus amyloliquefaciens, Curtobacterium pusillum ou Saccharomyces cerevisiae. Modulation of cell endogenous membrane potential by an external electrical field influences the structure and function of membrane compartments, proteins and lipid bi-layer. In this work, the effects of applied potential on Saccharomyces cerevisiae growth were characterized through simple yet conclusive experiments. Cell growth time profile and cell division were investigated as macroscopic response to the electrical stimulation. Control experiments were conducted under identical conditions except for the absence of applied potential. An ecological study on Saccharomyces cerevisiae populations in spontaneous fermentation has been conducted in three vats of a cachaça distillery in Minas Gerais, Brazil. Ninety-seven yeast isolates were collected at the beginning, the middle and at the end of the production period, and were identified by standard methods. Differentiation between the indigenous S. cerevisiae strains isolated was performed by mitochondrial DNA (mtDNA) restriction analysis, RAPD-PCR, and PCR fingerprint using an intron splice primer. Saccharomyces cerevisiae HAU-1, a time tested industrial yeast possesses most of the desirable fermentation characteristics like fast growth and fermentation rate, osmotolerance, high ethanol tolerance, ability to ferment molasses, and to ferment at elevated temperatures etc. However, this yeast was found to be sensitive against the killer strains of Saccharomyces cerevisiae. Aflatoxins are hepatotoxic metabolites produced by Aspergillus flavus and A. parasiticus on a number of agricultural commodities. This research was carried out to evaluate the ability of thermolysed and active Saccharomyces cerevisiae to attenuate liver damage caused by aflatoxin. Diets were prepared containing 0 aflatoxin; 400 mug kg-1 aflatoxin; 400 mug kg-1 aflatoxin plus 1% of dehydrated active yeast, and 400 mug kg-1 aflatoxin plus 1% of thermolysed yeast. Cells of yeasts adhered in corn grains and the storage perspective for use as probiotic BABT

Yeasts were applied to corn grains, containing 16 or 20% moisture, at concentrations of 1 and 2%. The cellular viability was assayed at 0, 15, 30, 90 and 110 days. The cellular viability did not differ statistically among the treatments up to 30 days of storage, with the median viability of 89.10%. The average viability found at 90 days (72.20%) was lower than in the first three storage periods. After 110 days, the average viability was 61.14%. In conclusion, since yeast cells were viable up to 110 days in storage on corn grains, these can be used as a vehicle for the application of yeast as a probiotic.

Os efeitos da parede celular de Saccharomyces cerevisiae (PCSc) na dieta para frangos foram avaliados com base nos tratamentos: dieta referência com avilamicina (AV); dieta referência (DR); DR com 0,1% de PCSc (PCSc0,1); DR com 0,2% de PCSc (PCSc0,2); DR com 0,3% de PCSc (PCSc0,3). O delineamento foi o de blocos ao acaso, com cinco tratamentos e seis repetições de 10 aves, totalizando 300 frangos, machos. Effect of probiotics and Yucca schidigera extract supplementation on broiler meat quality Animal Sciences

The current study investigated the effect of dietary supplementation with probiotics and Yucca schidigera extract on physicochemical parameters, proximate composition, mineral content and fatty acid profile of broiler breast and thigh muscles. In total, 240 one-day old broilers were randomly allocated into two dietary treatments groups: 1) Control (basal diet), 2) experimental (basal diet with two probiotics Pediococcus acidilactici and Saccharomyces cerevisiae and Yucca schidigera extract). The results showed that the pH value was higher in the experimental group than in the control group (p < 0.05). Dietary yeast supplementation may improve the digestive efficiency of ruminants, but responses depend on the yeast strain and the diet composition. Corn silage and citrus pulp are usual carbohydrate sources for dairy cows in southeast Brazil. This study evaluated the supplementation of dairy cows fedding on corn silage-citrus pulp-based diets with Saccharomyces cerevisiae CNCM I-1077 (Lallemand SAS, Toulouse, France). Twenty multiparous, midlactation Holstein cows were assigned to two treatments in crossover design.
